Hepatoprotective Role for Querectin in Diabetic Rats: Hypolipidemic and Antioxidant Effects, SALAH O. BASHIR

 

Abstract
Dyslipidemia and liver damage are common complications of diabetes mellitus (DM), therefore methods of their preven-tion are important in the longterm mangement of diabetes mellitus. Quercetin, a plant flavenoid, has well documented hypoglycemic and antioxidant properties. This study was designed to investigate whether quercetin adminstration would protect againest dyslipidemia and liver damage in diabetic rats and to determine whether the effect is mediated through amilioration of oxidative stress. Sixty male Sprague-Dawley rats were distributed equally among 6 groups: (I) Control group (C), (II) Quercetin treated control group (CQ), (III) Diabetic group (D), (IV) Diabetic Insulin treated group (DI), (V) Diabetic quercetin treated group (DQ) and (VI) Diabetic Insulin and quercetin treated group (DIQ). At the end of the experimental period (8 weeks), blood samples were collected and used to determine the serum levels of glucose, HbA1c, liver enzymes: alanine aminotransferase (ALT), aspartate aminotransferase (AST), alkaline phosphatase (ALP), total cholesterol (TC), triglycerides (TG), low density lipoprotein cholesterol (LDL-c), high density lipoprotein cholesterol (HDL-c) and very low density lipoprotein cholesterol (vLDL-c). Liver homogenate levels of malondialdehyde (MDA), superoxide dismutase (SOD) and glutathione peroxidase (GPx) were also measured. Liver histopathological examination was conducted. Treatment with either insulin or quercetin alone resulted in partial normalization of the biochemical and histopathological diabetic changes. Complete normalization of all parameters was achived when insulin and querecitin were combined. The results suggest that querecitin exerts a hypolipidemic and hypatoprotective effect on diabetic rats through improvement of both glycemic control and oxidative stress defences.
